Preparing Sterile Water

  1. Wash each bottle with soap, rinse with tap water, then rinse with distilled water.
  2. Fill each bottle with distilled water. Close each bottle but leave the cap loose.
  3. Wrap each bottle in aluminum foil, and label as DI and date using permanent marker on autoclave tape.

Preparing Lab Equipment

  1. Wash each piece of lab equipment thoroughly; the autoclave only sterilizes, it will not clean.
  2. Wrap each piece of equipment in aluminum foil and label with date on autoclave tape.

Using Autoclave

  1. Place wrapped equipment or sterile water bottles on tray, and load tray into autoclave.
  2. Turn on autoclave, and check that the proper program is selected. For sterile water select the beaker icon, 121oC for 15 minutes.
  3. Close autoclave door and tighten as much as possible.
  4. If “add water” light is on, use jugs to the right of autoclave and add via port on top of autoclave.
  5. Press “start.” Record time, date and what is being autoclaved in autoclave log; sign your name.
